The woman, suffering from an ear pressure problem for years, needed a nasal rinse delivery system. She was unable to use any of the existing systems due to the discomfort and pain caused by them distributing the fluid to the back of her throat and out the other nostril, which also produces cross contamination of allergens or bacteria.

Solving the Brain Drug Delivery impedance Problem

As evident by the existence of few drugs that work for neurological and mental health disorders, the delivery of drugs to the brain is impeded by the presence of several formidable physiological barriers and systemic circulation side effects. Except for intranasal delivery, all other drug delivery systems used to delivery drugs to the brain, such as oral, injection, intravenous, or transdermal, are adversely affected by systemic circulation and are substantially impeded by the blood-brain barrier (BBB). In fact, the BBB blocks 98% of small molecule compounds and 100% of large molecule compounds. In the case of oral, which is by far the most common drug delivery route of administration, the drug will additionally be impeded by the first-pass effect in the liver.

One of the advantages of intranasal drug delivery is that the nasal anatomy provides several routes to achieve direct-to-brain drug delivery.

However, the target regions within the nasal cavity for direct routing to the brain cannot be effectively reached unless the delivery device is anatomically designed. The existing devices on the market deliver the drug in a straight line, so they provide no directional control, and they cannot defy the laws of physicsand bend the stream to overcome the array of anatomical barriers in the nose.



The other co-founders of Neosinus Health, Dr. Kashif Mazhar and Dr. Magda Pugh—ENT surgeons with more than six decades of combined clinical experience— witnessed this challenge first-hand while attempting to treat patients with existing nasal delivery systems. In response, the team immersed themselves in many rounds of product design, fluid distribution modelling, clinical testing, and user feedback sessions with patients. The hard work paid off in the form of a novel delivery device platform that could eliminate all known side effects and enable targeted drug delivery across all regions of nasal anatomy in a configurable manner. Today, the Neosinus platform is market validated, fully patented, and recognized as best-in-class. It is the only anatomically designed and configurable intranasal delivery platform, and because of their unique patent portfolio, the device can be combined with both patented or off-patent drugs and nanoparticle systems. To elucidate, many off-patent drugs have been around for decades and have a favorable safety profile, with potential to treat some of the greatest unmet clinical needs. Although traditional pharmaceutical companies pass them over because they are off-patent, Neosinus views them as valuable assets that can reduce the time to market and expand access through lower pricing.

Additionally, there are nanoparticle drug delivery enhancement systems, but intranasal delivery can only benefit from them if the drug can be delivered to the target region for optimal drug absorption and distribution within the nasal cavity.
